On the interaction of lignin peroxidase with lignin

چکیده
The mechanism by which lignin peroxidase (Lip) interacts with the lignin polymer is discussed. Veratryl alcohol (Valc), a secondary metabolite of white rot fungi, acts as a cofactor for the enzyme. The Lip-redox cycle is discussed in terms of Marcus theory of electron transfer. It is proposed that reaction of a nucleophile in the active site channel with the incipient Valc'. is an essential event in the cycle. Subsequently, outside the channel the Valc'. is regenerated and will oxidize the lignin polymer. Further it is postulated that a carbohydrate residue from an 0-glycosylation site at Ser-334 may act as the nucleophile in this process.

Determination of lignin-modifying enzymes (LMEs) in Hyphodermella species using biochemical and molecular techniques
White-rot basidiomycetes are one of the most important lignolytic microorganisms. These fungi have been reported to secrete three main classes of lignin degrading enzymes: lignin peroxidases (LiPs), manganese peroxidases (MnPs) and laccases. In this study, for the first time the lignin degrading capability of two plant pathogens i.e. Hyphodermella rosae and H. corrugata was evaluated using both...
متن کاملImprovement of catalytic performance of lignin peroxidase for the enhanced degradation of lignocellulose biomass based on the imbedded electron-relay in long-range electron transfer route
BACKGROUND Although lignin peroxidase is claimed as a key enzyme in enzyme-catalyzed lignin degradation, in vitro enzymatic degradation of lignin was not easily observed in lab-scale experiments. It implies that other factors may hinder the enzymatic degradation of lignin. Irreversible interaction between phenolic compound and lignin peroxidase was hypothesized when active enzyme could not be r...
متن کاملPostharvest Assessment of Lignifying Enzymes Activity, Flower Stem Lignification and Bending Disorder of Gerbera Cut Flower
Scape bending disorder is the most important factor affecting postharvest loss of gerbera cut flowers. One of the ultimate reasons for gerbera stem bending is lignin, with deformation structural functions and defensive mechanisms. This postharvest experiment was conducted to evaluate the role of phenylalanine ammonia lyase (PAL) and peroxidase (POD) enzymes activity in stem bending of two gerbe...
متن کاملDirect interaction of lignin and lignin peroxidase from Phanerochaete chrysosporium.
Binding properties of lignin peroxidase (LiP) from the basidiomycete Phanerochaete chrysosporium against a synthetic lignin (dehydrogenated polymerizate, DHP) were studied with a resonant mirror biosensor. Among several ligninolytic enzymes, only LiP specifically binds to DHP. Kinetic analysis revealed that the binding was reversible, and that the dissociation equilibrium constant was 330 micro...
متن کاملPolymerization of monolignols by redox shuttle-mediated enzymatic oxidation: a new model in lignin biosynthesis I.
Lignin is one of the most abundant biopolymers, and it has a complex racemic structure. It may be formed by a radical polymerization initiated by redox enzymes, but much remains unknown about the process, such as how molecules as large as enzymes can generate the compact structure of the lignified plant cell wall. We have synthesized lignin oligomers according to a new concept, in which peroxid...
